Ethylene Glycol Bis(sulfosuccinimidylsuccinate) (Sulfo-EGS) is a crosslinking agent used in biological research to study peptides and proteins. Each end of the Sulfo EGS molecule contains Sulfo-NHS esters (N-hydroxysuccinimide esters with a sulfonate group), which react with primary amine groups on polypeptide chains and the side-chain of lysine amino acids. The two NHS esters are separated by a 12-atom spacer arm.
Sulfo-EGS is water-soluble and membrane-impermeable, making it ideal for labeling the surfaces of cellular membranes. The crosslinks formed by Ethylene Glycol Bis(sulfosuccinimidylsuccinate) can be broken by reacting with hydroxylamine at a pH of 8.5.
A related compound, EGS (Ethylene Glycol Bis(succinimidylsuccinate)), is water-insoluble, which makes it membrane-permeable. EGS is used primarily for studying intracellular interactions.
